Inrecentyears,microfluidicshavebroughtgreatconviniencetocellseparation,thereforelargenumbersofpeopledevotethemselvestostudymethodsofcellisolation.PinchedFlowFractionation,duetoitssimplemanipulationandhighapplicationscope,hasdrawnresearchscholars'attention,sinceintroduced.
Inthispaper,wearegoingtoemployimmersedboundary-latticeBoltzmannmethodtostudyPinchedFlowFractionation,andlatticeBoltzmannmethodisutilizedtoconstructfluidfieldandimmersedboundarymethodisusedtosimulatetheinteractionbetweencellsandfluid.
Firstly,themodelandmethodsemploiedinthispaperarevalidatedcarefully.
Thevalidationarebasedonthreeaspects:(1)validationoflid-drivencavityflow;(2)validationoftwo-dimensionalPoiseuilleflow;(3)validationofflowaroundacircularcylinder.Bycomparingthesimulationalresultswithpreviousresults,itiscanbeconcludedthatthemethodsandmodelscansimulatethefluidflowaccurately.
Moreover,theseparationperformancesofdifferentconditionsarestudied.TheseparationeffectsatlowReynoldsnumber(about1)ofdifferentpinchedsegmentwidths,flowratios,andcellmembranestiffnessarediscussed,andthecellbandingpheonomenonisinvestigated.
Theconclusionsarefollowing:
(1)Thepinchedsegmentwidthcaninfluencetheisolationperformancetremendously,Inmostcases,decreasingthepincedsegmentwidthcanimprovetheseparationperformances.
(2)Inordertoseparatetwokindsofcells,thedilutedflowratemustbemorethanthatofparticlesolution.Enlargingthedilutedflowratecanenchancetheseparationperformances.However,whenthedilutedflowrateincreasestosomevalue,itisnoteffectiveanymoretoimprovetheisolationeffects.
(3)Theempiricalformulacanpredicttheoutflowpostionsofcells.Butwhencellsaretooflexible,theformulawillbenotsoeffective.
(4)Whenthedistancebetweentwocellsaretoosmall,cellbandingwilloccur.
Andalso,therelativepositionbetweentwocellswillinfluencethesepartioneffectaswell.Withtheincreaseofdistancebetweentwocells,approximateisolationresultsareobtainedwhensmallcellisinthefront,backandleftpositionoflargecell.However,abetterseparationperformancecanbegotwhensmallcellisintherightpostionoflargecell.
